NEW WARNING
Millennium: The Takeda Oncology Company issued a "Dear Health Care Professional" letter concerning updates to prescribing information for bortezomib (Velcade). Bortezomib is indicated for treatment of multiple myeloma and mantle cell lymphoma in patients who have received at least one prior therapy. The new prescribing information includes updated survival data in patients with previously untreated multiple myeloma, which is 36.7 months. Bortezomib is metabolized by liver enzymes; exposure to the drug is increased in patients with moderate to severe hepatic impairment. Therefore a dosage modification has been added for patients with moderate to severe hepatic impairment. For these patients, initially begin with 0.7 mg/m2 during the first cycle, increase to 1 mg/m2 for subsequent doses, or reduce to 0.5 mg/m2 if the higher dose isn't tolerated. Monitor the patient closely for signs of toxicity.
